Design of Targeted and Release Controlled Liposome for Paclitaxel and Doxorubicin Combination in Breast Cancer Therapy

Abstract

Paclitaxel and doxorubicin are commonly used in chemotherapy of breast cancer. Now we have developed a kind of nanocarrier liposome that can deliver the combination of Dox and PTX to alleviate the pain brought by side effects and decrease the resistance. In this paper, we expected the result data of the drug loading capacity and the drug loading efficiency, the release of PTX and Dox in different pH environment, and the absorption of drugs which is estimated by the amount of free drug remains in the cells. The combination of Dox and PTX in liposome was modified with folic acid for tumor targeting, and achieved pH responsive drug release in tumor cell by introduction of N-(4-carboxybenzyl)-N, N-dimethyl-2,3-bis (oleoyloxy) propan-1-aminium (DOBAQ), a kind of pH sensitive lipid. The double layers were loaded with hydrophobic drug PTX, and the hydrophilic drug DOX is loaded in the aqueous core of the vesicle. We expected the results to reduce side effects and improve specificity when treating the cancer.
