The Effect of Moderate Intensity Exercise and Folate Acid on Homocysteine level in Rattus Novergicus Hyperhomocysteine

Abstract

Introduction: Hyperhomocysteine is an increased plasma level of total homocysteine. In humans, the level of homocysteine is regulated by diet, age, gender, genetics, drugs, renal function and lifestyle. The terminology homocysteine does not reflect a pathological condition but represents a biochemical abnormality with direct consequences in the variation of the pathological condition. High levels of homocysteine are one indicator of cardiovascular risk (Lentz et al., 2004). Every increase of 2.5 umol of plasma homocysteine is related to increased stroke risk (Clarke et al., 2004). The purpose of this study was to determine whether a decrease of homocysteine in the treatment group given folate acid, moderate intensity exercise and combination of folate acid and moderate intensity exercise. Methods: This research was a true laboratory experiment with a randomized post test control group design, using replication and a control group for comparation. Twenty-eight Rattus Norvegicus at 3 months of age were randomly allocated into 4 groups, group K control group (n=7), group P1 folate diet 2 mg/kg/6 weeks (n=7), group P2 moderate intensity exercise training 5 times/week/30 minutes/6 weeks (n=7), and group P3 combination folate diet and moderate intensity training (n=7). Before starting, the experimental subjects were given methionine with a dose of 1 mg/kg/ day for 4 weeks. Blood samples were collected 24 hours after last exercise training, and the ELISA method was used for examination of homocysteine levels. Result: Statistic analysis used ANOVA, which indicated that there was no significant difference between group treatment and group control. Conclusions: There is no significant difference with moderate intensity exercise and folic acid due to several reasons, perhaps not adequate amounts of folic acid, and in this study before research there was no examination of folic acid level. Until now there is no exactly clear formula of exercise such as duration, intensity, frequency or volume exercise.
