Abstract

The pomegranate fruit has many benefits for human’s health and often used as traditional medicine. Pomegranate seeds are rich in antioxidant and antibacterial, but the active substances were not yet known so a test should be done to determine it. The extract obtained through maceration process was crude extract that contains several compounds, so it should be purified through fractionation i.e. the compounds separation process based on the degree of polarity. The aim of this study was to identify the active substances in the fraction of pomegranate seeds ethanolic extract. Laboratory experimental through Liquid-liquid fractionation using Hexan, Ethyl Acetate and Ethanol as solvents, continue with Thin Layer Chromatography on Kiesel gel plate GF 254 with Hexan: Ethyl Acetate: Formic acid as the developers, UV 254 lamp and Column Chromatography against the Hexan fraction with silica gel 60 static phase, a 2.5 cm in diameter column glass and 25 cm height of absorbent was done. Grouping of compounds determined by the visible spots. Based on the results of Thin Layer Chromatography, there were 4 groups of compound according to the visible spots. As the conclusion, there were 4 groups of compound in the red pomegranate seed ethanolic extracts.
