Abstract

Background: The treatment of alveolar bone defects and juvenile periodontitis with the use of a bone graft still has limitations. The concept of tissue engineering using Stem Cells from Human exfoliated Deciduous teeth (SHED) with high potential of proliferation and hydroxyapatite scaffold (HA) with osteoconductive properties as excellent biomaterials can be an alternative for bone regeneration in patients with such cases. Purpose: To observe BMP-2 expression as an indicator of bone regeneration after SHED application on a hydroxyapatite scaffold. Methods: This research was a laboratory experimental study with a post-test only control group design. SHED implantation with a hydroxyapatite scaffold was placed on a tooth socket of 8 male wistar rats after anterior mandibular tooth extraction for 21 days. Immunohistochemical examination was performed to ascertain the number of osteoblasts and BMP-2 expression. Results: BMP-2 in the group of hydroxyapatite scaffold without SHED expressed 7.25 osteoblasts while in the group of hydroxyapatite scaffold with SHED expressed 12.5 osteoblasts. Statistical analysis of an independent T-test showed p = 0.016 < 0.05 so that there was a significant difference between the number of osteoblasts expressing BMP-2 in the group of hydroxyapatite scaffold without SHED and in the group of hydroxyapatite scaffold with SHED. Conclusion: The expression of Bone Morphogenic Protein 2 (BMP-2) after application of a hydroxyapatite scaffold with SHED was higher than a hydroxyapatite scaffold without SHED.
