Abstract: Kedungwuni Timur Subdistrict in Pekalongan Regency is an endemic DHF region with the increase of cases in the last three years. Vector control using insecticide is an effective way to break the transmission, whether done by the government or individuals. It has known that using too much insecticide for long time can cause resistance of mosquito that is indicated by increasing the Monooxygenase activity. The aim of this study was to measure resistance status of Aedes aegypti from endemic rural in Pekalongan Regency using Monooxygenase assay. The research was conducted in May- June 2017 with a cross-sectional design in 4 endemic villages in Kedungwuni Timur, Pekalongan Regency. Sample of Larva were collected using a dipping technique. Mosquito homogenate was stored and used for biochemical assays to identify Monooxygenase enzyme activity. The result showed as much as 70% mosquitos in Kedungwuni Timur Subdistrict had been resistant to Pyrethroid insecticide with an average value of abso rbance 1.18630. These results also showed that there was no different resistance status among 4 research sites (ANOVA, p = 0,476). Resistance to synthetic Pyrethroid insecticide in Aedes aegypti population in Kedungwuni Timur Subdistrict might be caused by the mechanism of detoxification enzymes in particular Monooxygenase.
