The Effect of Physical Exercise on the Astrocytes of the Cerebellar Cortex Exposed to Monosodium Glutamate in Rattus Norvegicus

Abstract

Background: Monosodium glutamate (MSG) is known as the most popular food additive used worldwide. Several studies have reported the neurotoxicity effect of MSG leading to astrocytes apoptosis in the cerebellar cortex. It is believed that excessive MSG correlates with the reactive oxygen species (ROS) accumulation that activates the p38 signaling pathway resulting in cell apoptosis. Physical exercise is reported to have anti-oxidant, anti-inflammation, immunomodulator and metabolic effects. The neuroprotection and neuroplasticity effect of physical exercise is also reported through the increase of brain-derived neurotrophic factor (BDNF) concentration. Objective: To evaluate the effect of physical exercise on the number of astrocytes in the cerebellar cortex of exposed to MSG in Rattus norvegicus. Design: Experimental study. Subject: 28 Rattus norvegicus strain Wistar (male sex, 6-8 weeks old, weight between 100-150 g). Intervention: MSG exposure at a dosage of 6 g/kg once daily and treadmill exercise. Outcome: The number of astrocytes in the Purkinje layer. Results: There were significant differences (p <0.05) in the mean number of astrocytes between the control group and the MSG group, between the MSG group and the MSG combined with treadmill group and between the treadmill group and the MSG combined with treadmill group. No significant difference (p> 0.05) was observed between the treadmill group and the control group and between the MSG combined with treadmill group and the control group. Conclusion: MSG administration caused marked reduction in the number of astrocytes in the cerebellar cortex. The provision of treadmill exercise inhibits the effect of MSG on astrocytes.
